This project investigated the skin's permeability to the analgesic drug, buprenorphine hydrochloride. The aim of the work was to measure the penetration of the drug through human epidermal membrane in vitro as a function of pH by radiochemical and HPLC techniques. The effects of chemical penetration enhancers, azone and oleyl alcohol, and electrical enhancement (iontophoresis), on skin permeability to buprenorphine hydrochloride were also investigated. The distribution, or partition, of the drug between stratum corneum and aqueous buffer was studied and compared with that for the organic solvents octanol, butanol and isopropyl myristate. The permeability coefficients of buprenorphine hydrochloride in epidermal membrane were largely unaffected by changes in the pH of the applied drug solution; the range of values was ....
